Vinpocetine chemical sensor for its dissolution testing, assay and as HPLC detector

Abstract

An electrochemical detector for vinpocetine (Vp) in its pure state and pharmaceutical dosage forms was prepared based on the ion associate of vinpocetine with phosphotungstic acid. The sensor was fully characterized in terms of its membrane composition, life span, effect of soaking, pH, interferences and temperature and exhibited a Nernestian response of (59.6 ±0.6 mV/decade) in the concentration range 1.0×10-2-10.0 ×10-5 M. The sensor was successfully applied to the plotting of dissolution profile study of Vp capsules and tablets and the results were validated by comparison with spectrophotometric assays, according to the quality control unit of the capsules and tablets pharmaceutical company producers. Also, Vp was assayed potentiometrically in pure solutions and in pharmaceutical preparations by using the developed electrode either under batch or flow injection conditions with recovery values ranged from 98.50 to 101.65%, and with relative standard deviation of 0.21 to 0.89%. The method was also applied to the determination of vinpocetine in its pure form under HPLC conditions ranged from 1.0 × 10-4-10.0 × 10-6 M. The presented chemical sensor for vinpocetine is developed to be used in quality control and quality assurance during manufacture of vinpocetine drug and pharmaceutical preparations. The applications of this sensor were also extended to monitoring the dissolution profile of the drug, as well as to potentiometric electrochemical detection of the drug under HPLC conditions, which does not consist a common application of such kind of chemical sensors.
